Under which of the following conditions will there be no change in the reading frame of following mRNA? 5' AACAGCGGUGCUAUU 3'
- Insertion of A and G at 4th and 5th positions respectively
- Deletion of GGU from 7th, 8th and 9th positions
- Insertion of G at 5th position
- Deletion of G from 5th position
Answer
(2) Deletion of GGU from 7th, 8th and 9th positions
Solution
The codons are read in triplets without punctuation:
5' AAC AGC GGU GCU AUU 3'
Bases 7, 8 and 9 form the third codon GGU.
Deleting these three bases removes one whole codon:
5' AAC AGC GCU AUU 3' — the remaining codons are read unchanged.
As per NCERT, insertion or deletion of one or two bases changes the reading frame from the point of change (frameshift), whereas insertion or deletion of three bases (or multiples of three) inserts or deletes one or more codons and the reading frame from that point remains unaltered.
Options (1), (3) and (4) (one or two bases) cause frameshifts.
