The figure below shows three steps (A, B, C) of Polymerase Chain Reaction (PCR). Select the option giving correct identification together with what it represents ? [figure]

  1. C - Extension in the presence of heat stable DNA polymerase
  2. B - Denaturation at a temperature of about 98 separating the two DNA strands
  3. A - Denaturation at a temperature of about 50
  4. A - Annealing with two sets of primers

Answer

(1) C - Extension in the presence of heat stable DNA polymerase

Solution

In the figure: A = the double-stranded DNA (template) with the region to be amplified; B = single strands with short primers (black) bound — annealing; C = primers extended along the whole region — extension.

As per NCERT, in extension the primers are extended using the nucleotides and the thermostable DNA polymerase (isolated from Thermus aquaticus, Taq polymerase).

Denaturation (about 94–98 ) separates the strands; it is not shown as B and does not occur at 50 ; A is not annealing.

∴ C – Extension in the presence of heat stable DNA polymerase.
